Mulla on ehkä vähän erilainen tarina miten mun "ura" lähti käyntiin. Tein ammattikorkeakoulussa (Haaga-Helia, tietojenkäsittely) viimeisillä kursseilla huvikseni omaa sivustoa, koska nämä luennot eivät hirveemmin kiinnostaneet. Samalla tietenkin mietiskelin, että milläs tästä opinnäytetyöstä selvitään helpoimmalla kun ei kirjoittaminen todellakaan ole mun juttu. No keksin että hei, kehitän mobiilipelin sillä yksi vaihtoehto toiminnallisesta opinnäytetyöstä oli "oman liiketoiminnan aloittaminen". Yleensähän opinnäytetyöt ovat perinteisiä tutkimuksia, portfolioita koulussa tehdyistä töistä tai jollekkin yritykselle ratkaisun toteuttaminen.
Helmikuussa 2018 peliprojektini alkaessa aiemmin mainittu sivusto oli kerännyt sen verran mainostuloja, että nämä ulos saamiseksi oli pakko perustaa yritys. Tästähän aina vitsailen että vahinkolapsen lisäksi olen vahinkoyrittäjä.

Pelissä ei hirveästi sisältöä ollut enkä odottanutkaan tältä muuta kuin opinnäytetyön läpipääsyä. Eikä oikeastaan selvää suunnitelmaa ollut ammattikorkeakoulun jälkeen sivustoni kehittämisen lisäksi, eikä sitä nyt aina tarvitsekkaan miettiä liikoja eteenpäin.
Peli ei alkuun luonnollisestikkaan menestynyt, mutta koronan aikana eli noin 2 vuotta myöhemmin Streamer Sim Tycoon pääsi Google Play kaupan "Suositellut"-osioon muutamissa maissa ja keräsi kasaan miljoona latausta. Nyt peli valittiin top 20 finalisteihin Huawein Apps UP EU kilpailussa!

Varmasti tuli todella "epäammattimaista" tekstiä, mutta tämä oli mun oma nöyrä ja rehellinen tarinani ilman turhaa mahtailua! :)

Ob es bei Kindern schädlich ist, keine Ahnung. Kommt auf die Vielfalt an, ebenso wie bei der "normalen" Ernährung. Wenn man sich falsch ernährt (Vitamine auslassen, zuviel von einer Art, etc.), ist jede Form falsch.
Bei Tieren wiederrum ist die fleischlose Ernährung definitiv ungesund. Der Hund ist zu 100 % ein Fleischfresser und benötigt das auch. Die Reißzähne sind nicht ohne Grund bei Hund und Katze vorhanden.Damit wollte die Natur die nicht beim reißen von Grünzeug helfen, sondern bei Fleisch.
Ebenso ist der Organismus von einem Tier anders und benötigt andere Stoffe zum Leben.Beim Menschen kann man hier mit Ersatzprodukten helfen. Beim Tier bitte nicht!

Der Hund wurde bereits Minuten nach der Geburt mit tierischen Produkten gefüttert und zwar von der Mutter mit der Milch. Klar gibt es hier Ersatzprodukte, aber darauf sollte man wenn es geht verzichten.
Das ist mit Absicht so überspitzt, denn manchmal kommen mir die Argumente so weit hergeholt vor, das dann auch sowas berücksichtigt werden muss.

Leste en artikkel om at SARS-CoV-2 er et RNA-virus som muterer ofte. Så også her at det er en variant av virus i Trondheim nå som er mere smittsom enn tidligere.
Det jeg lurer på er om dette da er samme "art" som den som oppsto første gangen; eller er det en ny art? Når vil det i så fall bli en ny art?
Så vidt jeg har forstått så er COVID-19 navnet på sykdommen som er forsaket av SARS-CoV-2 viruset så hvor mye kan viruset mutere før det blir en "ny" sykdom?

Unlike current conventional MSC manufacturing processes applied by other companies in this sector, Cynata Therapeutics (Cynata) took a different approach:
- evaluating possible shortcomings / bottlenecks of existing processes on their way to at-scale manufacturing down the track (past Phase 3 clinical trials),
- looking for an alternative,
- creating a platform,
- testing it01107-X/fulltext),
- validating its at-scale manufacturing capability,
- then taking your product/s to the clinic.
Cynata's Cymerus™ manufacturing platform utilises induced pluripotent stem cells (adult cells reprogrammed to an embryonic stem cell-like state, maintaining the essential properties of embryonic stem cells, but without the ethical and legal concerns surrounding them).
These induced pluripotent stem cells (iPSCs) are turned into a (mesoderm-derived) precursor cell known as mesenchymoangioblast (MCA), before generating the final Cymerus™ iPSC-MSCs. Source: Cynata Investor Presentation 14 August 2020 The manufacturing process consists of three stages:
- iPSC banking,
- iPSC expansion and differentiation to MSCs to freeze and intermediate bank and
- MSC expansion and formulation of final clinical product.
The above outlined cutting-edge technology eventually resulted in the world's first clinical trial of an allogeneic iPSC-derived MSC product, evaluating the safety and efficacy of Cynata's lead product CYP-001 for the treatment of 15 patients with steroid-resistent acute graft-versus-host disease (SR-aGvHD) with very encouraging results: "The Phase 1 clinical trial enrolled 15 patients with steroid-resistant acute GvHD. The primary evaluation at Day 100 revealed highly promising safety and efficacy results, as announced on 18 December 2018. By Day 100, the Complete Response and Overall Response rates were 53% and 87% respectively, and overall survival was at least 87%. Participants who completed the Primary Evaluation Period then continued to a follow-up period for up to two years after the initial dose, which has now been completed. The overall survival rate after two years was 60% (9/15 patients), which compares favourably with previously published outcomes. An overall survival rate of just 17% after two years has been reported in patients with steroid-resistant acute GvHD who received standard of care treatment.1 Furthermore, a recently published review article summarised numerous studies with bone marrow and adipose tissue derived MSCs in patients with steroid-resistant acute GvHD. Six of the studies reviewed reported two year survival in MSC-treated patients, which ranged from 16.6% to 40%.2 Similarly, in another recently published Phase 3 trial of an investigational drug (ruxolitinib) in patients with steroid-resistant acute GvHD, overall survival after 12 months was 38%, while there were insufficient surviving patients remaining on study to calculate the survival rate at later timepoints.3 The results of Cynata’s trial illustrate the significant potential of CYP-001 as a new treatment option for GvHD."
In order to further validate its Cymerus-MSCs by showing that the cells have similar or superior functionality to native, donor-derived MSCs, Cynata is building a large catalogue of pre-clinical data covering numerous diseases

A look under the hood Based on the existing banking strategy, approximately 9 × 10^4 vials, each containing 1 × 10^6 iPSCs, can be generated from a single iPSC line. At the current processing scale, a single vial of 1 × 10^6 iPSCs is capable of giving rise to 3.2 × 10^10 MSCs on average, while the entire iPSC bank has the capacity to generate 2.9 × 10^15 MSCs, or 29 million clinical doses (each containing 1 × 10^8 MSCs).
Compared to other companies that have stated to be capable of producing enough clinical doses to treat between 500 to 20,000 patients from the starting material from just one donor,
Cynata's Cymerus™ platform would allow to overcome a number of important limitations, including
Conversely, since iPSCs can proliferate indefinitely, and MCAs themselves can expand into extremely large quantities of MSCs, Cynata expects to be able to manufacture all of the cells that it will ever need from a single Master Cell Bank of iPSCs – derived from a single donor.
- dependence upon donors
- variability between donors
- the relative scarcity of MSCs in adult tissue
- the low proliferative capacity of adult stem cells
- loss of potency and reduced efficacy associated with extensive MSC culture expansion (in an effort to maximise the number of MSCs from each donation).

iPSC = Cancer?
Cynata has also thought of a way to avoid the teratoma formation risk that is associated with (undifferentiated) iPSCs in vivo by incorporating the following steps to the manufacturing process:
- after the induction of mesoderm, cells were cultured in a single-cell suspension in semisolid medium, which does not support iPSC survival;
- cells were passed through a mesh filtration step, which eliminates small clumps of undifferentiated iPSCs; and
- iPSC-derived MSCs were expanded in adherent cell culture conditions, which do not support survival or expansion of undifferentiated iPSCs.

Where does the iPSC starting material come from?
The iPSC starting material is currently being provided by FujiFilm Cellular Dynamics (which was used in Cynata's World-First clinical trial of an allogeneic iPSC-derived therapy). The original license agreement was entered into with Cellular Dynamics back in September 2014

Professor James Thomson and his team at the University of Wisconsin - Madison, including Professor Igor Slukvin (one of the founders of Cynata) were one of two independent research groups that first reported the creation of iPSCs from human cells (along with Professor Shinya Yamanaka et al, at Kyoto University, Japan).
On 30 March 2015 however, CDI was acquired by FujiFilm for an amount of US$307 million
, representing a premium of 108% to CDI’s closing price on 27 March 2015.FujiFilm now also happens to be the licensee of Cynata's lead product CYP-001 in GvHD
.Having said that, Cynata's Cymerus™ iPSC-MSC platform could also be utilised using iPSC starting material from other companies, which also makes it a very lucrative approach for companies that already have access to their own iPSC line and/or are looking into genome editing.
It's all about the patents
The iPSC step is covered under the license agreement between iPS Academia and (now) FujiFilm Cellular Dynamic (FCDI).
A license agreement with the Wisconsin Alumni Research Foundation (WARF), which is the licensing and patenting atm of the University of Wisconson, Madison, where Professor Igor Slukvin MD, PhD, has developed this technology, granted Cynata world-wide exclusive rights to certain patented (one granted patent and certain patent applications) IP forming part of the Cynata Technology comprised in Appendix B-2 ["Generation of clonal mesenchymal progenitors and mesenchymal stem cell lines under serum-free conditions"] the License Agreement as described in Section 8 in relation to making, using, selling, importing, and offering for sale certain products for certain therapeutic and diagnostic uses [...]
The original Cynata Technology has since been further developed, with parts of the original process being eliminated and replaced by licensed progressive improvements. This commercially viable and improved Cymerus™ manufacturing platform, now capable of generating MSCs with superior performance compared to the original process, is covered by an Estate of patents both worldwide exclusively and non-exclusively licensed to Cynata as well as patents fully owned by Cynata.
For example, while early IP covered production of MCAs from PSCs using OP9 Feeder cells changed to MSCs to MCAs (still using mouse feeder layers), there is now exclusively licenced IP covering production of MSCs from MCAs and MCAs from PSCs under defined conditions in the absence of non-human components (Tenascin-C). One of them being this one here: "Methods and materials for hematoendothelial differentiation of human pluripotent stem cells under defined conditions"
(patent family: US14206778 & patent application US20140273211A1), which is as per ASX announcement dated 7 December 2017
owned by WARF and to Cynata.
Cynata filed for a patent (amongst others) citing this one, called "Colony forming medium and use thereof"
, inventors being Igor Slukvin, Gene Uenishi, Derek Hei and Diana Drier.
This patent Estate is best described by saying that the core technology has been licensed from WARF with own patents (not licensed from WARF) around it. Whilst the core patent 7,615,374 has an anticipated expiration of 1 Februart 2028, other patents covering improvements that, unless being utilised won't result in the same final Cymerus-MSC product, have anticipated expiration as far as 24 January 2035

covering this very topic in regards to mobile phones used today, stating the following:"This is particularly important when it comes to filing new patent applications, as each new ‘inventive step’ can be patented in its own right, allowing many different aspects of an invention to be protected. Securing multiple patents can also mean that even if an earlier patent expires, the latest developments added to the product will continue to be safeguarded from being plagiarised."
Regarding a very vague, therefore in my opinion a last resort desperate attempt to start a "composition of matter" argument, I would strongly recommend to check out the following links:
Although investors of other stem cell companies regulalry refer to possible patent infringements due to their belief that one company can actually "own" MSCs full stop and although there is no guarantee, I am very confident that all the parties listed above have done their due dilligence over the years to have this patent Estate secured.
Licensing - isn't that expensive? If the Company exercises its rights to sub-license certain rights under the License Agreement, it must pay US$10,000 to WARF and also 30% of any fees received by Cynata from such sub-licensees in addition to certain annual and milestone fees, certain royalty and non-royalty payments as well as pay WARF certain reimbursement of patent fees and costs.
Whilst this might sound a bit "juicy", you may want to consider the alternative to Cynata's conservative approach of paying fees (US$10,000 per sub-license, 30% on milestone payments plus single digit royalties on net sales) on money it has actually received and is sitting in its bank account:Outright purchase
of a / part/s of a platform or development of a new platform
from scratch, requiring ongoing Capital Raises
resulting in dilution of existing investors and / or loan/s
(perhaps with - uncertain - future income to be used to service these loans and / or additional shares to be issued to repay the capital and therefore also resulting in further dilution of existing investors).
The above alternative/s could be compared with negative gearing, hoping that the income generated from this platform will eventually outweigh the ongoing expenses including interest and (lower) royalty payments (as there is generally at least one patented component required for the manufacturing process that needs to be in-licensed) and in this instance, will also allow for the loan/s to be repaid down the track with whatever is left.
My personal view on investing aligns with the conservative approach taken by the company. Having said that, Cynata is still considered to be a speculative, high-risk investment, especially in the absence of an (ongoing) income stream sufficient to cover ongoing expenses.

So... I have a dog name Marvim and he is very calm and sweet... With most people. Even when strangers come in the house he likes them. Most of the times (like 70%), when my mom picks or touches him, he growls at her.
Sometimes just slighty, some others are really bad and he even exposes his teeth and gums to her. Sometimes I can see it's just he playing. Like when he sees her moving her hands a lot (he likes to ""bite"" her hands), but others he looks angry for real. He won't even let my mom brush him anymore, because he starts to do this and tries to escape. But that doesn't happens with only brushing, can be as simple as my mom trying to pet him or toching him.
Also, when my mom comes near Marvim's toys when he is playing, he starts to treat to bite her, agressively, but when I do the same he is okay with it. He even likes to play lying in my lap. Someone can help with this?
He did not bite her for realin all this time heis being with us (as far as I know), so could be nothing, but this is really odd and my mom is complaining about this behavior. He always did this, but now its happening away too often.
I tried to look at Google for some help, but I couldn't find anything about such a especific behavior.

So I thought instead of just posting my trades
, I would also explain my thought process so you can see how I use the various indicators to make decisions.
• 9:31am – buy of SU at $14.90. The all out dump this morning was overnight bots reacting to Trump virus news. First 5 minutes are always peak fear. Got in when the 5 minute RSI was done at 15, the 15 minute was at 20, and both outside of the BB
• 10:25am – sell of all SU shares at $15.38. My list says 9:51am but that’s because I always put in a ‘fake’ sell order. So I had the shares limit sell at $42 and then I just readjust when I want to sell. I sold in this area
. Thought process was that we just ran from LOD to HOD on straight green bars and just got to moving averages so we were likely due for a short term pullback.
• 10:43am – buy back all position + few extra scalped shares at $15.25. See graph above and you will see 2 big red bars but on little volume. Scalping 13 cents on 2,500 shares is $325 saved.
• 12:43pm – sold my SU position for $15.85 here on the 5 minute timeframe
. I rebought back $15.77 the next 5 minute candle as it back tested the moving average.
This is basically how I’m going to play SU going forward. Scalp when I see opportunities and then I’ll save half US cash for trading. You can look at what I did today as three different things:
- You can either just lower your cost average (I would have kept same shares but lowered by cost average by $0.21);
- Add shares to my position (my first sell was for 2502 shares and last buy back was for 2525 shares); or
- Just think of the profit - $0.21 cents saved on 2500 shares is $500ish after fees. This is a technique my buddy taught me when he was in a tough position. Couldn’t sell because it was too big of a loss, didn’t want to throw money at the problem, so he scalped for shares where he could.

Chart Perspective – Most things probably had same chart today. Futures and everything dropped on Trump virus news. This is what happens when BOTS read headlines but then when markets open and people can react, you usually get a buying of the dip. Basically what we saw on SPY and most of the names in this sector. I'm developing. It's free (unless you want to pay me :P), and supports language learners by quickly enabling them to look up words in online dictoinaries either frame in frame (iframe), or via a popup/tab, when highlighting a word (and or hitting a hotkey).

I currently support two german-english dictionaries as well as a german-chinese one, and many other languages. If you have any wishes or criticisms let me know. I'm working on this for free as a hobby, and am of course wishing for many people to be helped out by it.

2020.09.25 05:43 only_for_hentai The hilariously screwed up order in which I watched this show.
I have no shame in admitting that the main factor that got me to watch this show was the fact that Yin looks vaguely like Rei from Evangelion (....and that it was apparently critically acclaimed and highly related, but who cares about that, amirite?), so I kind of went in with only a very basic Wikipedia summary of the show's premise. So I looked for a stream on Google and clicked the first link I could find, not knowing that I'd actually started watching Season 2. (I also assumed it had 12, and not 24, episodes, because that's the episode count I saw when I started watching).
This in and of itself isn't THAT bad, but four episodes in I decided I wasn't happy with the quality of the stream and switched sites... this time clicking on the proper season, *starting at Episode 5*, because that's where I thought I'd left off.
Thing is, I'd heard that the show has shorter episodic arcs, so the fact that the story shifted in a completely different direction didn't really stick out as a red flag. This really wasn't helped by the fact that Eps 5 and 6 of season 1 involve Havoc, who really looks a LOT like the two kids from Season 2, so I assumed it was an extended flashback arc showing Hei's relationship with (who I assumed was their mother). It wasn't until eps 7 and 8 (the first detective story) that I thought something might've been off.
At that point I decided to go back to Episode 1 on the new site I'd been watching on and realized I recalled nothing, which is when I realized I'd made a really dumb mistake and started properly watching it from the beginning and having a much better time with it.
Dunno how this is particularly relevant to anything, but Darker than Black is probably the worst I've ever fucked up the physical act of watching a show.

Hi all, I'm hoping someone can help me out or point me in the right direction.
I've got a file (135kb, 11s) that I'm trying to get a certain "noise" out of. I was able to capture a noise my fish makes but there's a ton of background noise also (water, pump, etc) so I was hoping someone could help me isolate it OR point me in the right direction on a way to DIY or if neither is possible, methods for getting pro help.
I've tried the basic sound editors in the Google play store and what I've found available for pc via a quick search and none were able to do what I'm looking for. But I'm also a major noob and could simply be looking for the wrong thing or doing it the wrong way.

I vividly remember these sweet candies that used to exist when I was growing up in the 2000s, that would look exactly like these
erasers that you would on the tip of a pencil. The candies were to scale, so you could also put them on top of a pencil.
They were hard, sweet candy in the same vein as SweetTarts. None of my friends seem to remember them, but my sister does. There’s absolutely no evidence of them on Google, though. I’m dying to remember what they were called.

2020.09.15 18:59 Kerudo Desktop Client Font Fix
After playing a while on Android I decided to try and play on my laptop and immediately noticed that all the "normal" fonts were rendering with Times New Roman and everything was ugly, and some things like the % indicator in loadings was even misaligned. Overall everything looked out of place.
I thought I'd try to fix that so I reverse engineered the APK and uncompressed the Unity asset bundles that were holding the fonts. I found a bunch, including the nice brush fonts used in the menus etc.
Only one font that was sans serif was called SimHei so I installed it and tried the client again, and bingo!

My guess is that the devs didn't realize that the SimHei font isn't by default on all OSes. I think it's a font available through some Windows feature to install additional fonts, but is totally missing from macOS. Searching "SimHei font" on Google returns a bunch of download links so you might be lucky there.

TLDR; installing font "SimHei" fixes the game and makes it look like it does on mobile

First, the result: Fenix 6s Sport loop
This post will describe the steps I took to add a sport loop to the Garmin Fenix 6S pro solar. These steps however can be applied to all Fenix 6S series though.
Coming from an apple watch with a sport loop like this one
, I wanted the same type of strap on my Garmin as this strap has the lowest amount of pressure points on your wrist and generally feels a lot nicer compared to the default strap in my opinion.
After some googling, I found the following aftermarket strap: https://nl.aliexpress.com/item/33015080566.html
. The problem here however is with the connector. This strap uses the quick fit connector and an older spring loaded type of connector. As it’s using both, the amount of rigid non-nylon parts are extended compared to just using the first part (connected to the strap) and this causes discomfort on my wrist as the plastic bits still dig into your skin.
The solution here is to use the following type of sport loop: https://nl.aliexpress.com/item/33008116479.html
. This loop is basically the previous loop minus the quick-fit connector. As you can see from the description, this doesn’t natively fit with the Fenix 6(s) as the Fenix series has pins which are a lot ‘beefier’ compared the the pins that came with the aftermarket strap. The strap can be modified with a couple of simple steps:

- The first thing you want to do is slightly enlarge the pin holes in the aftermarket strap. This can be done by using a hex key (size 1.5mm) which is slightly larger compared to the hole in the aftermarket strap and try to gently push it in while turning at the same time. Doing so, you’ll slightly enlarge the hole until the original Fenix 6s pin fits into the aftermarket strap as shown in the picture here:

- The next problem consists around being unable to retract the pins anymore (the aftermarket pins come with a kind of lever). This is solved by filing a little bit of the material on the section with the hole on the strap. Doing so, you can retract the original Fenix pins with a sharp object as shown in the picture here:
The result? A directly connected sport loop that skips the use of the quick fit connector making it a bit lighter and a LOT more comfortable!
It’s a lot of finicky work to fix this properly but knowing that you’ll probably wear this watch 24/7, it better be comfortable 😅. Hope this reference helps anyone in the future with the same problem, feel free to PM me if you need any help with this solution!
